Description

YmeCas12a is derived from a Yellowstone metagenome and is a thermostable, crRNA‑guided DNA endonuclease. It exhibits strong catalytic activity across a broad temperature range (37–60 °C), with an optimal reaction temperature of 56 °C. YmeCas12a preferentially recognizes the PAM sequence TTTV (V = A/C/G). For double‑stranded DNA (dsDNA) targets, efficient recognition and cleavage require the presence of a PAM site, whereas single‑stranded DNA (ssDNA) targets can be cleaved without PAM dependence.

In addition, both dsDNA and ssDNA targets can activate the trans‑cleavage activity of YmeCas12a (also referred to as collateral or auxiliary cleavage). Once YmeCas12a binds with crRNA and the target DNA to form a ternary complex, the enzyme becomes activated and nonspecifically cleaves surrounding ssDNA reporters in the reaction mixture. Because of these properties, YmeCas12a can be used not only for specific in vitro cleavage of dsDNA but also for rapid detection of target nucleic acids.

Furthermore, it should be noted that YmeCas12a exhibits a length preference for ssDNA reporter probes.
