TtAgo Endonuclease originates from recombinant E. coli and contains the cloned Argonaute (Ago) nucleases gene from Thermus thermophilus. It is a nucleic acid endonuclease that relies on guide DNA sequences to complementarily pair with target nucleic acids, thereby specifically recognizing and cleaving them. Under the guidance of 5’-phosphorylated guide DNA (>15 nt), TtAgo Endonuclease precisely cleaves single-stranded DNA substrates. The cleavage site is located between the 10th and 11th nucleotides starting from the 5’ end that is complementary to the guide sequence, and this cleavage is not restricted by adjacent sequences of the target site.
