Description
SpCas9 is a DNA endonuclease derived from Streptococcus pyogenes, guided by crRNA and tracrRNA (or a single sgRNA). It specifically recognizes and cleaves double-stranded DNA (dsDNA) targets in a PAM (NGG)-dependent manner. Cas9 introduces a double-strand break at a site located 3 bp upstream of the PAM sequence. In addition, SpCas9 can specifically cleave single-stranded DNA (ssDNA) or single-stranded RNA (ssRNA) in the presence of a DNA PAMmer sequence. SpCas9 is also widely used in vitro for targeted DNA cleavage and cloning applications.
This product is a glycerol-free, lyophilized SpCas9 nuclease that can be stored and transported at ambient temperature. It is available in multiple lyophilized formats (powder or bead) to meet the requirements of different downstream applications.

Reconstitution Recommendation
Dissolve 100 μg of lyophilized SpCas9 in 124 μL of reconstitution solution to obtain a final enzyme concentration of 5 μM (5 pmol/μL).
Storage
- SpCas9 Nuclease (Lyophilized) + Cas Reconstitution Solution: Stable at room temperature for storage and transport. After reconstitution, store at –20 °C and avoid repeated freeze–thaw cycles.
- 10× Buffer for Cas9: Store at –20 °C; transport at 4 °C.
Notes
- To prevent RNase contamination, keep the work area clean and tidy. Always wear clean gloves and a mask during operation, and ensure that all consumables (pipette tips, centrifuge tubes, etc.) are RNase-free.
- This product contains only the Cas nuclease. Target DNA and crRNA required for the CRISPR reaction system must be prepared separately. For the 10× Cas9 buffer, it is recommended to source from TOLOBIO.
- After reconstitution, SpCas9 should be handled on ice when preparing reaction mixtures. Immediately return the enzyme to –20 °C after use. At –20 °C, the reconstituted enzyme is stable for up to one month.
