Description
Cas9 Nickase (H840A) NLS, also known as Cas9 Nickase (H840A) NLS, is a nuclease with a Nuclear Localization Signal (NLS) that specifically cleaves single-stranded DNA under the guidance of gRNA. Cas9 Nickase (H840A) NLS is an H840A mutant created by a single amino acid mutation in the HNH domain of the Cas9 nuclease. Like the wild-type Cas9 nuclease, Cas9 Nickase (H840A) NLS targets the DNA sequence with the help of gRNA. However, due to the mutation in the HNH domain, Cas9 Nickase (H840A) NLS only cleaves the DNA strand non-complementary to the gRNA.
Features
- This product has a low off-target rate and high specificity. Gene editing requires two gRNA targets (generally 0-20bp apart), and the double-strand cleavage produces 5' ends, facilitating homologous recombination. This greatly reduces off-target effects, enhancing gene-editing specificity and avoiding negative impacts from non-specific editing in subsequent experiments.
- With nuclear localization signals, the editing efficiency is high. Cas9 Nickase (H840A) NLS contains nuclear localization signals (NLS) of SV40 T antigen at both the N-terminus and C-terminus of the protein, allowing the Cas9 Nickase and gRNA complex to quickly enter the nucleus from the cytoplasm after transfection, significantly increasing gene-editing efficiency. Cas9 Nickase can enter cells via microinjection, electroporation, and liposome-mediated methods, and this DNA-free system eliminates the risk of foreign DNA integration into the cell genome.

In vitro screening of efficient and specific gRNA; in vivo gene editing using electroporation or microinjection with two specific gRNAs.
Purity
Purity ≥95% after SDS-PAGE electrophoresis; free from DNA exonucleases, non-gRNA dependent DNA endonucleases, RNAase, and phospholipase.
Storage
Store at -20ºC, effective for at least one year. For longer storage, aliquot and store at -80ºC, avoiding repeated freeze-thaw cycles.
Precautions
- When using this product, ensure RNase-free and DNase-free conditions for gRNA and DNA handling. All self-prepared reagents and consumables should be Nuclease-free. If there is potential nuclease contamination, consider treating overnight with 0.01% DEPC, followed by high-temperature sterilization. It's recommended to wear a disposable mask during operation.
- Add RNase Inhibitor to the reaction system to protect RNA from degradation.
