Description

TccCas13a is a crRNA‑guided RNA endonuclease derived from Thermoclostridium caenicola. Compared with commonly used Cas13a orthologs such as LwaCas13a and LbuCas13a, TccCas13a exhibits markedly enhanced thermostability. In its RNP form, TccCas13a maintains strong catalytic activity across 42–72 °C, with an optimal reaction temperature of 56–60 °C. Guided by crRNA, TccCas13a specifically recognizes single‑stranded RNA (ssRNA) targets and does not strictly depend on a protospacer flanking site (PFS).

TccCas13a also displays robust trans‑cleavage activity (also referred to as collateral or auxiliary cleavage). Once TccCas13a binds crRNA and the target RNA to form a ternary complex, the enzyme becomes activated and nonspecifically cleaves surrounding ssRNA molecules in the reaction mixture. This property has been widely leveraged for the development of rapid nucleic acid detection assays.

In addition, TccCas13a exhibits a distinctive cleavage preference toward certain ssRNA reporter probe designs.
