spCas9 is a DNA endonuclease guided by a crRNA and tracrRNA complex (or a fused sgRNA) derived from the S. pyogenes strain. In the presence of a PAM sequence (5'-NGG-3') on the target double-stranded DNA, it specifically cleaves the target DNA, causing double-strand breaks and generating blunt ends. The PAM is essential for both the recognition and cleavage by Cas9, with the cleavage site located within the target sequence, three bases away from the PAM region. Additionally, in the presence of a DNA PAMmer sequence, SpCas9 can also specifically cleave single-stranded DNA or RNA. This enzyme is also applicable in experiments such as in vitro target DNA cleavage and cloning of desired fragments.

Description
