Description
Cas12a nuclease (formerly known as Cpf1) is an RNA-guided endonuclease that specifically cleaves target double-stranded DNA in the presence of a PAM sequence. Unlike Cas9, Cas12a has several distinct features: (1) It requires only a single crRNA, is smaller in size, and is more easily delivered into cells; (2) It produces sticky ends upon cleavage, which are more conducive to precise genome editing; (3) Its cleavage site is distant from its recognition site, allowing for the possibility of consecutive editing; (4) It recognizes different PAM sequences compared to Cas9, offering alternative editing site options. Different Cas12a proteins require different PAM sequences for target recognition, with some recognizing TTN sites and others recognizing TTTN sites.
