SuCas12a2 nuclease is derived from Sulfuricurvum sp. PC08-66. Cas12a2 is a crRNA-guided RNA endonuclease whose direct repeat (DR) sequence is highly similar to that of Cas12a. In the presence of a protospacer flanking site (PFS) adjacent to the target single-stranded RNA (ssRNA), Cas12a2 specifically recognizes and cleaves the target ssRNA. Compared with other Cas nucleases that, once activated by their cognate targets, exhibit collateral cleavage activity limited to ssDNA (e.g., Cas12a, Cas12b), ssRNA (e.g., Cas13a), or both ssRNA and ssDNA (e.g., Cas12g), Cas12a2 demonstrates a much stronger and broader trans-cleavage activity. Upon formation of the ternary complex with crRNA and target RNA, Cas12a2 is activated to indiscriminately degrade non-specific nucleic acids—including ssDNA, ssRNA, and dsDNA—fragmenting all such molecules present in the reaction system.

Description

SuCas12a2 nuclease is derived from Sulfuricurvum sp. PC08-66. Cas12a2 is a crRNA-guided RNA endonuclease whose direct repeat (DR) sequence is highly similar to that of Cas12a. This product is a glycerol-free, lyophilized SuCas12a2 nuclease, enabling storage and transport at ambient temperature. It is available in multiple lyophilized formats, including powder and bead forms, to accommodate diverse downstream assay requirements.

Reconstitution Recommendation 

Dissolve one vial of 100 µg lyophilized SuCas12a2 in 139 µL of reconstitution solution. The resulting enzyme concentration will be 5 µM (equivalent to 5 pmol/µL).

 

Storage

  • SuCas12a2 Nuclease (Lyophilized) + Cas Reconstitution Solution: Stable at room temperature for storage and transport. After reconstitution, store at –20 °C and avoid repeated freeze–thaw cycles.
  • 10× Buffer for SuCas12a2: Store at –20 °C; transport at 4 °C.

 

Notes

  • To prevent RNase contamination, keep the work area clean and tidy. Always wear clean gloves and a mask during operation, and ensure that all consumables (pipette tips, centrifuge tubes, etc.) are RNase-free.
  • This product contains only the Cas nuclease. Target DNA and crRNA required for the CRISPR reaction system must be prepared separately.
  • After reconstitution, SuCas12a2 should be handled on ice when preparing reaction mixtures. Immediately return the enzyme to –20 °C after use. At –20 °C, the reconstituted enzyme is stable for up to one month.
