Description
PfAgo, derived from recombinant expression in Escherichia coli, is a programmable DNA endonuclease. Under the guidance of a short 16-18 nucleotides of 5'-phosphorylated single-stranded DNA (gDNA), the corresponding specific sequence on the substrate is digested, and a break is introduced in the phosphodiester main chain of the complementary substrate sequence.
The advantage of PfAgo is that the guiding design and target sequence selection are not limited by PAM, and it has high activity on single-stranded DNA and most double-stranded DNA substrates. In the presence of Mg2+ or Mn2+ metal ions, the enzyme obtains RNase H like activity and cuts the substrate at the base complementary to the 10th and 11th bases of the DNA guide chain.
