Description

SaCas9 is a CRISPR nuclease derived from Staphylococcus aureus and is one of the most widely used compact Cas9 orthologs, offering distinct advantages for therapeutic genome editing. The protein consists of 1,053 amino acids with a molecular weight of approximately 120 kDa—about 25% smaller than SpCas9 (1,368 aa). This reduced size makes SaCas9 particularly well‑suited for delivery platforms with strict cargo limitations, such as AAV vectors.

SaCas9 recognizes the 5'-NNGRRT-3' PAM sequence (R = A or G), which is longer and more complex than the NGG PAM recognized by SpCas9. The increased PAM specificity theoretically enhances targeting precision by reducing the number of potential off‑target sites across the genome. Experimental studies have shown that SaCas9 exhibits strong editing activity in mammalian cells, with cutting efficiencies comparable to—or in some cases exceeding—those of SpCas9.

A unique enzymatic feature of SaCas9 is its multi‑turnover activity. Unlike SpCas9, which typically functions as a single‑turnover nuclease, SaCas9 can bind and cleave multiple DNA substrates sequentially under certain conditions. This property may contribute to improved editing efficiency in scenarios where nuclease concentration or delivery efficiency is limited.

  • AAV‑Mediated In Vivo Genome Editing: Its compact size enables packaging into a single AAV vector, making SaCas9 highly suitable for in vivo therapeutic delivery.
  • Gene Therapy Applications: Widely used in AAV‑based treatments for ocular disorders, neurodegenerative diseases, and other conditions requiring efficient in‑tissue editing.
  • Tissue‑Specific Editing: Achieves targeted genome modification when combined with AAV serotypes exhibiting distinct tissue tropisms.
  • Dual‑Nickase Strategies: SaCas9 D10A/N580A nickase variants can be paired to generate high‑fidelity double‑nick editing with reduced off‑target activity.
  • Orthogonal Genome Editing: Functions alongside SpCas9 to enable multiplexed or dual‑gene editing without crosstalk between systems.
