Description

MS2‑p65‑HSF1‑mCherry is a key auxiliary component of the Synergistic Activation Mediator (SAM) CRISPRa system. It builds upon the MS2‑p65‑HSF1 activation module by incorporating the red fluorescent protein mCherry, enabling simultaneous potent transcriptional activation and real‑time visualization.

The fusion protein consists of three functional elements:

  • MS2 coat protein, which specifically recognizes MS2 aptamer hairpins engineered into the sgRNA scaffold.

  • p65 and HSF1 activation domains, two strong transcriptional activators that synergistically recruit co‑activators and transcriptional machinery to drive robust gene upregulation.

  • mCherry, a bright and photostable red fluorescent reporter with minimal spectral overlap with GFP, allowing multiplexed imaging and straightforward tracking of protein localization and expression dynamics.

Together, these components allow MS2‑p65‑HSF1‑mCherry to amplify CRISPRa‑mediated transcriptional activation while providing a built‑in optical readout for monitoring delivery, expression, and subcellular distribution.
