Description
FnCas12a (also known as Cpf1) nuclease is derived from the strain Francisella tularensis. FnCas12a is a crRNA-guided DNA endonuclease. In the presence of a PAM (TTN) sequence on the target double-stranded DNA, it specifically cleaves the target double-stranded DNA, inducing double-strand breaks with sticky ends. In contrast, the specific cleavage of single-stranded DNA targets by FnCas12a is PAM-independent. Once FnCas12a assembles into a ternary complex with crRNA and target DNA, it triggers collateral cleavage of non-specific single-stranded DNA (ssDNA), degrading any ssDNA sequences present in the reaction system. For this reason, FnCas12a can be applied for rapid detection of target nucleic acids.
This product is glycerol-free lyophilized FnCas12a nuclease, which allows transportation and storage at ambient temperature. It is available in multiple formats including lyophilized powder and lyophilized beads to accommodate diverse application scenarios.
Components
- FnCas12a Nuclease (Lyophilized)
- Cas Reconstitution Solution
- 10× Buffer for Cas12a
Reconstitution Recommendation
Dissolve 100 μg of lyophilized FnCas12a in 130 μL of reconstitution solution to obtain a final enzyme concentration of 5 μM (5 pmol/μL).
Storage
- FnCas12a Nuclease (Lyophilized) + Cas Reconstitution Solution: Stable at room temperature for storage and transport. After reconstitution, store at –20 °C and avoid repeated freeze–thaw cycles.
- 10× Buffer for FnCas12a: Store at –20 °C; transport at 4 °C.
