Description
sssLbCas12a (single-stranded DNA-specific LbCas12a) is a mutant engineered from wild-type LbCas12a endonuclease. Obtained via targeted engineering and screening, this mutant specifically recognizes target ssDNA while exhibiting extremely low activity for the recognition and cleavage of target dsDNA. Similar to the mechanism of action of wild-type LbCas12a, sssLbCas12a is crRNA-guided to recognize and cleave target ssDNA in a PAM-independent manner. In addition, recognition and binding to target ssDNA can activate the trans-cleavage activity of sssLbCas12a, degrading ssDNA probes present in the reaction system. Therefore, sssLbCas12a can be applied for rapid detection of target nucleic acids.
