Aflatoxins are toxic metabolites produced by certain fungi such as Aspergillus flavus and Aspergillus parasiticus. They are highly carcinogenic and are primarily found in grains, peanuts, nuts, cottonseed, animal feed, vegetable oils, as well as in animal tissues and blood. Among them, aflatoxin B1 exhibits the highest toxicity, carcinogenicity, and contamination frequency. Aflatoxin B1 Immunoaffinity Column selectively adsorbs aflatoxin B1 from the sample solution, thereby providing highly targeted purification of the sample. The sample solution after column purification can be directly used for HPLC or LC-MS/MS analysis.

Among them, aflatoxin B1 exhibits the highest toxicity, carcinogenicity, and contamination frequency.

Aflatoxin B1 Immunoaffinity Column selectively adsorbs aflatoxin B1 from the sample solution, thereby providing highly targeted purification of the sample. The sample solution after column purification can be directly used for HPLC or LC-MS/MS analysis.

 

Principle

The basis of the assay is the antigen-antibody reaction, where antibodies are immobilized within the column. The sample, containing aflatoxin B1, undergoes extraction, filtration, and dilution. Subsequently, it slowly passes through the aflatoxin B1 immunoaffinity column, where the toxin binds to the antibodies inside the column. Afterward, the immunoaffinity column is washed to remove any unbound impurities. Aflatoxin B1 is then eluted with methanol and injected into the analytical instrument for detection.

 

Precautions

  • Before use, the immunoaffinity column must be brought back to room temperature (22-25°C).
  • Store the immunoaffinity column at 2-8°C, do not freeze.
  • Please use the immunoaffinity column within its expiry date.
  • Sample volume: Adjust the sample volume accordingly, either increasing or decreasing as needed, and ensure that the volume of the extraction liquid is adjusted accordingly.
  • Column capacity: 300ng. If the concentration of the toxin in the sample, divided by the dilution factor, exceeds the column capacity, adjust the volume of the sample appropriately and retest.
  • pH: The pH of the sample solution applied to the immunoaffinity column should be between 6 and 8. If it deviates from this range, adjust the pH with hydrochloric acid or sodium hydroxide.
  • Maintaining consistency between the solvent used during analysis and the mobile phase can eliminate solvent effects.
  • Aflatoxin B1 is carcinogenic. Protective gear such as gloves and masks should be worn during handling.
  • Containers and solutions containing aflatoxin B1 that have been used are best soaked overnight in a 5% sodium hypochlorite solution.
