Description

PrimeIAmp™ HotStart Bst P DNA/RNA LHNB LyoMix, meticulously developed by SBS Genetech, is compatible with both DNA and RNA target detection. Built on the rapid and robust Bst P polymerase, it offers stable, fast, and highly sensitive amplification. The incorporation of HotStart technology further reduces false positives. Its proprietary lyophilized formulation enhances versatility—supporting routine research and enabling seamless lyophilized product development and manufacturing with a unified reagent platform.

Product Features:

  1. HotStart Aptamer-Controlled Enzyme Activation: HotStart Bst P includes a thermostable aptamer that inhibits >90% of enzyme activity below 30 °C, with complete activation achieved within 1 minute above 60 °C. This supports room temperature setup and significantly minimizes nonspecific amplification at lower temperatures.

  2. High-Temperature Operation: The reaction temperature of 70 °C reduces primer-dimer formation, enhances amplification specificity, and improves nucleic acid release from crude samples.

  3. Helicase-Enabled Prime LAMP: The presence of helicase enables pLAMP (prime LAMP) amplification without requiring F3/B3 outer primers and permits halving the input of FIP/BIP primers. This configuration further reduces nonspecific amplification and significantly improves amplification uniformity.

The reagent includes LHNB dye (Leuco-HNB), which initially forms a deep violet complex with Mg²⁺ prior to amplification. As the LAMP reaction progresses, pyrophosphate (PPi) produced during amplification binds Mg²⁺, leading to the release of LHNB and a visible color change from deep violet to pale green.

Storage

For long-term storage, keep the product at –20 °C or below (stable for up to 18 months). The reagent can withstand up to 10 freeze–thaw cycles without performance loss, though repeated freeze–thawing should be avoided when possible. For short-term use, storage at 2–8 °C is recommended, where the product remains stable for up to 2 weeks. Due to the high concentration of lyophilization excipients, a small amount of white insoluble material may be observed upon thawing; this will fully dissolve after 3–5 minutes at 30 °C (or by warming in the palm), without affecting product performance.

 

Precautions

  • The recommended reaction temperature for Bst P polymerase in LAMP amplification is 70 °C. Since the helicase component also operates optimally at 70 °C, performance is significantly reduced at 65 °C or lower.
  • This product contains a high concentration of salts. Appropriate personal protective equipment (PPE) should be used to avoid contact with skin, eyes, nose, and respiratory tract. In case of accidental exposure or inhalation, rinse thoroughly with plenty of water.
  • To prevent aerosol contamination, operations should be performed in separated, designated areas wherever possible.
