Description

PrimeIAmp™ HotStart Bst DNA/RNA LHNB LyoMix, meticulously developed by SBS Genetech, is tailored for LAMP amplification and supports both DNA and RNA target detection. Based on a fast and highly tolerant Bst DNA/RNA polymerase, it achieves stable, rapid, and highly sensitive amplification. The integration of HotStart technology further reduces the risk of false positives.

Its proprietary lyophilized formulation grants the reagent exceptional versatility—fulfilling routine research needs while also streamlining product development and large-scale lyophilized reagent manufacturing with a unified formulation.

Product Features: HotStart Bst DNA/RNA polymerase is equipped with a heat-activated aptamer that ensures >90% inhibition of enzyme activity below 30 °C and complete activation within 1 minute above 60 °C. This design facilitates room temperature setup and significantly minimizes nonspecific amplification at lower temperatures.

The reagent also contains LHNB (Leuco-HNB), a chromogenic dye that initially forms a deep violet complex with Mg²⁺. As the LAMP reaction proceeds, pyrophosphate (PPi) generated during amplification binds Mg²⁺, leading to the release of free LHNB and a visible color change to pale green.

Storage

  • For long-term storage, keep the product at –20 °C or below (stable for up to 18 months). The reagent can withstand up to 10 freeze–thaw cycles without performance loss, though repeated freeze–thawing should be avoided when possible. For short-term use, storage at 2–8 °C is recommended, where the product remains stable for up to 2 weeks. Due to the high concentration of lyophilization excipients, a small amount of white insoluble material may be observed upon thawing; this will fully dissolve after 3–5 minutes at 30 °C (or by warming in the palm), without affecting product performance.
  • SYBR Green dye is photosensitive and prone to degradation. Protect from light during storage and use. The recommended final concentration is 0.75×.

 

Precautions

  • This product contains a high concentration of salts. Appropriate personal protective equipment (PPE) should be worn to avoid contact with skin, eyes, nose, and respiratory tract. In case of accidental contact or inhalation, rinse immediately with plenty of water.
  • To minimize the risk of aerosol contamination, operations should be conducted in separate designated areas whenever possible.
