Description
The DP‑Probe (Displaceable Probe) is a strand‑displacement fluorescent probe designed specifically for fluorescence‑based LAMP amplification. When used together with HotStart Bst P Basic reagents, the Loop DP‑Probe delivers exceptional specificity and significantly reduces nonspecific amplification.
The Loop DP‑Probe Pair is generated by modifying one of the Loop primers (either LF or LB). It consists of two annealed, labeled oligonucleotides:
a quencher probe (5’ IBFQ + Tail + Loop), and
a reporter probe (Tail‑complementary region + 3’ fluorophore).
The annealed pair is non‑fluorescent. During the LAMP reaction, the Loop primer invades the “dumbbell” structure, extends during amplification, and displaces the reporter probe, which is released into solution and accumulates as a fluorescent signal.
