Description
PrimeIAmp™ HotStart Bst P DNA/RNA Basic Microbead is a fully formulated lyophilized microsphere, containing HotStart Bst P DNA/RNA polymerase, helicase, dNTPs, Mg²⁺, reaction buffer salts, and lyophilization excipients. Each microbead supports a 20 µL amplification system.
Performance Characteristics of Bst P:
HotStart Control: Bst P is equipped with a heat-activated aptamer, ensuring >90% enzyme inhibition at temperatures below 30 °C and full activity release within 1 minute above 60 °C. This facilitates room-temperature setup and significantly reduces nonspecific amplification under low-temperature conditions.
High-Temperature Amplification (up to 70 °C): Elevated reaction temperatures effectively suppress primer dimer formation, improve specificity, and enhance nucleic acid release from crude samples.
Helicase-Assisted Prime LAMP: The inclusion of helicase enables pLAMP (prime LAMP) amplification without requiring F3/B3 outer primers. It also allows the input of FIP/BIP primers to be reduced by half, further minimizing nonspecific amplification and improving amplification uniformity.
This multi-purpose reagent is suitable for LAMP-based detection using Molecular Beacon probes, DP-LAMP probes, and lateral flow strip formats.

Storage:
- Store at –20 °C in a dry, sealed condition for up to 24 months. Alternatively, it can be stored at room temperature (25 °C) in a sealed container for up to 3 months.
- As the product is in a lyophilized form and highly hygroscopic, it is recommended to store it at –20 °C for up to 12 months after opening.
Precautions
- The recommended reaction temperature for Bst P polymerase in LAMP amplification is 70 °C. Since the helicase component also operates optimally at 70 °C, performance is significantly reduced at 65 °C or lower.
- This product contains a high concentration of salts. Appropriate personal protective equipment (PPE) should be used to avoid contact with skin, eyes, nose, and respiratory tract. In case of accidental exposure or inhalation, rinse thoroughly with plenty of water.
- To prevent aerosol contamination, operations should be performed in separated, designated areas wherever possible.
