Description
This product utilizes a chromatographic double‑antibody sandwich method to rapidly detect nucleic acid amplification products. Compared with traditional agarose gel electrophoresis, the nucleic acid test strip offers simple operation, rapid interpretation, is free of toxic substances, and requires no instruments or equipment.
For use, the user only needs to design primers/probes such that one is labeled with biotin (Biotin) and the other with fluorescein isothiocyanate (FITC) or 6‑carboxyfluorescein (6‑FAM). By ensuring that both labels are incorporated into the double‑stranded amplification product simultaneously, detection can be performed directly with this product.
Operating Steps
- According to the number of samples to be tested, take out the corresponding number of test strips and mark them on the absorbent pad (Figure 1). Each test strip can only be used once for a single sample.
- When the amplification product volume is 50–100 µL, nucleic acid products can be directly tested in a 200 µL PCR reaction tube. If the product volume is less than 50 µL, add ultrapure water to the PCR tube to make up the volume to 50 µL, mix thoroughly, and then proceed with detection.
- After PCR, RPA, or RAA reaction is completed, open the PCR reaction tube and insert the test strip with the binding pad end (arrow end) into the PCR tube (Figure 1). The liquid level must not exceed the upper edge of the binding pad. Wait until the interpretation area is fully saturated (approximately 1–2 minutes; in colder environments such as winter, absorption speed will be slower, and saturation time will be longer). Once the control line (C line) appears, the test strip can be removed.
- For RPA or RAA reactions, dilution is required before inserting the test strip.
- Observe the results within 10 minutes after the control line (C line) appears. Results read after 10 minutes are invalid.
- Record the test results, then seal and discard the test strip in a safe location.
Results
Positive (+):
A blue band appears at the control line (C line) and a red band appears at the test line (T line). A positive result indicates that the sample contains the target nucleic acid fragment, and its quantity is greater than the minimum detection limit of the test strip.
- When the concentration of the target nucleic acid product is low, the C line appears blue and the T line appears light red or even pale pink. This result should still be interpreted as positive.
- When the concentration of the target nucleic acid product is high, both the C line and the T line appear red. This result should also be interpreted as positive.
Negative (–):
A blue band appears at the control line (C line), while no band appears at the test line (T line). A negative result indicates that the sample does not contain the target nucleic acid fragment, or its quantity is below the minimum detection limit of the test strip.
Invalid:
No bands appear at either the control line (C line) or the test line (T line). This suggests that the test strip or amplification reagents may be damaged, expired, or that an operational error has occurred. In this case, carefully read the instructions, repeat amplification and testing. If the problem persists, immediately discontinue use of products from the same batch and contact your local supplier.
Precautions
- For research use only. This product is intended solely for scientific research. Before use, carefully read the instructions and strictly follow the operating procedures. Failure to comply may result in incorrect results.
- Storage and validity. Store the product according to the requirements stated in the instructions, under suitable environmental conditions and temperatures, and use it within the validity period. Improper storage or use of expired products may lead to erroneous results.
- Use promptly after opening. Once the package is opened, use the test strips as soon as possible to avoid moisture absorption, which may affect test performance.
- Environmental and operator factors. Insufficient lighting in the testing environment or operator color vision deficiencies may also cause incorrect results.
- Disposal and single-use notice. After use, promptly place the test strip into a sealed bag and dispose of it appropriately. This product is designed for one-time use only and must not be reused.
Storage
Store in a light-protected and dry place, at a temperature of 4~30°C, with a shelf life of 12 months.
