Description
This product utilizes a chromatographic double-antibody sandwich method to rapidly detect reporter molecules labeled with dual tags (FAM and Biotin or FITC and Biotin). Upon encountering target DNA/RNA, the Cas protein forms a functional complex with crRNA, which becomes activated and cleaves the reporter molecule completely. As a result, the test line (T-line) does not develop color.
Operating Steps
- Take out the appropriate number of test strips according to the number of samples. You may mark the cartridge to distinguish different experimental groups. Each test strip is for single use only.
- Add 2 μL of nucleic acid amplification product to 78 μL of Diluent Buffer and mix well. (This can be optimized based on specific experiments; 1–10 μL of reaction product can be diluted 9–200 times in a centrifuge tube using Diluent Buffer.)
- Apply 70 μL of the diluted reaction mixture to the sample well. Record the results in the interpretation area within 5 minutes.
- After recording the results, seal the test strip and discard it safely.
Result
- Negative: A visible band appears on the Test Line (T-line), and a band also appears on the Control Line (C-line). A negative result indicates that the target nucleic acid is either absent in the sample or present at a concentration below the detection limit of the test strip. (Cas protein is not activated, the reporter remains intact, and the T-line develops color.)
- Positive: No band appears on the Test Line (T-line), while a band is visible on the Control Line (C-line). A positive result indicates the presence of the target nucleic acid fragment in the sample. (Cas protein is activated, the reporter is cleaved, and the T-line does not develop color.)
- Invalid: No band appears on the Control Line (C-line), or both the Control Line (C-line) and Test Line (T-line) show no bands. This result suggests that the test strip is defective, damaged, or the procedure was performed incorrectly.

Storage
Test strips should be stored at room temperature. Avoid direct sunlight and moisture exposure.
Diluent Buffer should be stored at 2–8 °C.
