Description
This product is a lateral flow test strip featuring two bands: a lower Control Line (C-line) and an upper Test Line (T-line). Streptavidin (SA) is coated on the control line, while goat anti-mouse secondary antibodies are coated on the test line. Colloidal gold is conjugated with anti-FAM/FITC monoclonal antibodies.
The intact reporter molecule—labeled with Biotin on one end and FAM/FITC on the other—allows colloidal gold to be fully captured at the control line. When the reporter is cleaved by Cas enzymes, the colloidal gold bound to the cleaved fragments cannot be captured by the control line and instead forms a visible band at the test line.
The presence or absence of the test line indicates whether the Cas enzyme has been activated.
Reporter Design
In designing the reporter, one end is labeled with Fluorescein isothiocyanate (FITC) or 6-Carboxyfluorescein (6-FAM), and the other end is labeled with Biotin.
Operating Steps
- Take out the required number of test strips and label them accordingly.
- Transfer 10 μL of the CRISPR-Cas reaction product into a new PCR tube, then add 40 μL of ddH₂O and mix well. (If using a custom reaction system, it is recommended to optimize the dilution ratio.)
- Insert the test strip with the sample pad end immersed in the liquid. Let it sit at room temperature for 5–10 minutes, then interpret the results. (Ensure the liquid level does not exceed the “max” line.)
- After use, place the amplification product and test strip into a sealed bag and dispose of them properly.
Results
- Negative: No visible band on the Test Line (T-line), indicating that the reporter molecule was not cleaved and the Cas enzyme was not activated.
- Positive: A visible band appears on the Test Line (T-line), indicating that the nucleic acid probe was cleaved by the Cas enzyme and the enzyme was activated.
- Invalid: Neither the Control Line (C-line) nor the Test Line (T-line) shows a visible band, suggesting an operational error or that the test strip has degraded or malfunctioned. In this case, carefully review the instructions and repeat the test.

Storage
Test strips should be stored at room temperature. Avoid direct sunlight and moisture exposure.
