Description
Cas13a and crRNA form a functional complex that "slides" along the target nucleic acid sequence. Upon successful pairing, Cas13a is specifically activated, leading to the cleavage of surrounding reporter molecules. This kit combines isothermal amplification technology with Cas13a, offering high sensitivity, high specificity, and a high signal-to-noise ratio. It has been widely used in the field of molecular diagnostics for rapid and precise detection of pathogens.

- Nucleic acid test strips (FAM-Biotin product, we recommend Lateral Flow Assay for CRISPR SHERLOCK)
- Isothermal equipment (e.g., metal bath, water bath, PCR machine, etc.)
- Pipettes
- Nuclease-free water
- Target sequence-specific primers (for isothermal amplification)
- Note: Include T7 promoter in primer design: 5’-TAATACGACTCACTATAG-3’
- CrRNA/gRNA: Forms a functional complex with LwaCas13a and is specifically activated by the target sequence.
- LwaCas13a crRNA scaffold sequence: 5’-GAUUUAGACUACCCCAAAAACGAAGGGGACUAAAAC-3’
