Description
The DualHS G3mTTx qRT-PCR Lyophilized Bead contains nucleic acid aptamer–blocked hot-start G3Taq DNA polymerase, an Extreme ThermoStable Reverse Transcriptase (ET RTase), dNTPs, an Mg²⁺ activator, Mg²⁺, and lyophilization excipients. This formulation enables unbiased amplification of both DNA and RNA under Mg²⁺ conditions alone (no Mn²⁺ required). Designed specifically for TaqMan PCR (ROX-free), it provides highly sensitive detection of DNA and RNA molecules.
Thanks to the exceptional heat resistance of ET RTase, which remains fully active at 95 °C, both DNA and RNA viruses can be processed by simple heat treatment to release nucleic acids for subsequent PCR amplification. As a result, crude samples can be directly tested without nucleic acid purification.
Key Features
Dual Hot-Start Enzyme System
Both G3Taq and ET RTase are aptamer-blocked hot-start enzymes (activity suppressed >90% below 30 °C).
Enzyme activity is fully restored above 50 °C, ensuring precise regulation throughout PCR.
At the initial stage, a Blocker completely sequesters Mg²⁺, fully inhibiting polymerase activity. After heating at 95 °C for 2 min, the Activator hydrolyzes the Blocker, releasing Mg²⁺ and initiating amplification. This provides a dual hot-start mechanism.
Extreme Thermostability of ET RTase
ET RTase retains full activity even after heating at 95 °C for 15 min.
Through modification of the TTx enzyme’s ligand-binding core, its reverse transcription activity was shifted from Mn²⁺-dependence to Mg²⁺-dependence, enabling synchronized RT-PCR.
Optimized RT-PCR Performance
Maximum amplicon length from RNA templates: 500 bp.
Highest amplification efficiency: 70–150 bp.
Storage
Store sealed and dry at ≤ –20 °C: shelf life up to 60 months.
Store sealed and dry at room temperature: shelf life up to 12 months.
As the product is in lyophilized form and highly hygroscopic, once opened it is recommended to store at ≤ –20 °C, where it remains stable for 24 months.

The G3mTTx DNA/RNA qPCR Mix contains SBS Genetech’s engineered Extreme ThermoStable Reverse Transcriptase (ET RTase). This enzyme withstands 95 °C and efficiently reverse transcribes RNA up to 500 nt in length. Unlike conventional reverse transcriptases restricted to 37–50 °C, ET RTase enables cDNA synthesis at virtually any temperature, determined only by primer annealing conditions.
In addition, the system allows one-tube processing of crude samples—heat lysis, nucleic acid release, reverse transcription, and PCR amplification—streamlining workflows and redefining the format of molecular diagnostic reagents and instrumentation.
ET RTase is derived from a thermostable DNA polymerase and re-engineered using electronic scaffold reconstruction technology. Its ligand-binding core was modified to eliminate Mn²⁺ dependence, achieving strong reverse transcription activity with Mg²⁺ alone. This innovation removes the inhibitory effects of Mn²⁺ on PCR.
SBS Genetech provides two versions of the G3mTTx Mix:
CM Version – More flexible and user-friendly.
DualHS Version – Requires slightly stricter handling but delivers more rigorous amplification control.
Both formulations are glycerol-free. When combined with SBS Genetech’s proprietary 3M Spherical Lyophilization Excipient, they can be readily prepared into lyophilized beads containing amplification primers. This integrated design simplifies research, development, and production workflows while maintaining flexibility and reducing the complexity of reagent development and fixed-format manufacturing.
SBS Genetech also offers customized production of qPCR lyophilized beads, tailored to specific research or diagnostic needs.
