Description
5× DualHS G3mTTx qPCR Mix is a specialized 5× master mix for TaqMan-based Real-Time PCR, featuring dual aptamer-based HotStart enzymes—G3Taq DNA Polymerase and Extreme ThermoStable Reverse Transcriptase (ET RTase)—along with dNTPs, Mg²⁺ activator, Mg²⁺ ions, and stabilizers. The mix enables bias-free amplification of both DNA and RNA under Mg²⁺-dependent conditions (no Mn²⁺ required) and supports ultrasensitive detection of nucleic acids.
Due to the extreme heat resistance of ET RTase (retaining activity after 95°C treatment), the system allows direct detection of crude samples—thermal lysis efficiently releases nucleic acids from both DNA and RNA viruses, eliminating the need for nucleic acid purification prior to amplification.
Features
- Dual Aptamer-Based HotStart Enzymes: Both GSTaq and ET RTase are aptamer-blocked, maintaining >90% inactivity below 30°C and fully activating above 50°C. For experiments requiring enhanced HotStart stringency, a separate Blocker component can be added to sequester Mg²⁺ through insoluble complex formation. After heating at 95°C for 2 minutes, the Activator hydrolyzes the Blocker, releasing Mg²⁺ to initiate the reaction, achieving dual-layered HotStart control.
- Thermostable RT Performance: ET RTase maintains full reverse transcriptase activity after 15 minutes at 95°C. Its active center has been engineered to switch metal ion dependence from Mn²⁺ to Mg²⁺, enabling true one-step RT-PCR.
- Amplification Range: When using RNA templates, the system supports RT-PCR amplicons up to 500 bp, with optimal performance in the 70–150 bp range.
- Glycerol-Free and Lyophilization-Ready: The mix is free of glycerol and fully compatible with the proprietary 3M Spherical Lyophilization Excipient, simplifying the preparation of lyophilized PCR bead formulations. This integrated design streamlines research, development, and production, offering flexibility while reducing the challenges of reagent optimization and immobilization.

The G3mTTx DNA/RNA qPCR Mix contains SBS Genetech’s engineered Extreme ThermoStable Reverse Transcriptase (ET RTase). This enzyme withstands 95 °C and efficiently reverse transcribes RNA up to 500 nt in length. Unlike conventional reverse transcriptases restricted to 37–50 °C, ET RTase enables cDNA synthesis at virtually any temperature, determined only by primer annealing conditions.
In addition, the system allows one-tube processing of crude samples—heat lysis, nucleic acid release, reverse transcription, and PCR amplification—streamlining workflows and redefining the format of molecular diagnostic reagents and instrumentation.
ET RTase is derived from a thermostable DNA polymerase and re-engineered using electronic scaffold reconstruction technology. Its ligand-binding core was modified to eliminate Mn²⁺ dependence, achieving strong reverse transcription activity with Mg²⁺ alone. This innovation removes the inhibitory effects of Mn²⁺ on PCR.
SBS Genetech provides two versions of the G3mTTx Mix:
CM Version – More flexible and user-friendly.
DualHS Version – Requires slightly stricter handling but delivers more rigorous amplification control.
Both formulations are glycerol-free. When combined with SBS Genetech’s proprietary 3M Spherical Lyophilization Excipient, they can be readily prepared into lyophilized beads containing amplification primers. This integrated design simplifies research, development, and production workflows while maintaining flexibility and reducing the complexity of reagent development and fixed-format manufacturing.
SBS Genetech also offers customized production of qPCR lyophilized beads, tailored to specific research or diagnostic needs.
