Description
5× CM HS-G3mTTx qRT-PCR Mix (Lyo-Ready) is a specialized 5× master mix designed for TaqMan probe-based Real-Time PCR, containing chemically blocked HotStart CM HS-G3Taq DNA Polymerase, extremely thermostable reverse transcriptase (ET RTase), dNTPs, Mg²⁺, and stabilizers. The mix enables unbiased amplification of both DNA and RNA targets under magnesium-dependent conditions and supports high-sensitivity detection of nucleic acids from either source.
Thanks to the exceptional heat resistance of ET RTase, capable of withstanding 95°C, nucleic acids can be liberated from DNA or RNA viruses through thermal treatment—allowing direct detection from crude samples without nucleic acid purification.
Features
- HotStart Mechanism: The chemically modified CM HS-G3Taq DNA Polymerase remains fully inactive below 50°C and regains activity only after heating at 95°C for 5 minutes.
- Thermostable Reverse Transcription: ET RTase retains full activity even after 15 minutes at 95°C. Its active site has been engineered to shift metal ion dependence from Mn²⁺ to Mg²⁺, enabling synchronous RT-PCR in a single system.
- Optimized Probe Cleavage: Modification of the TTx polymerase active site reduces its 5'→3' exonuclease activity; the 3G Taq enzyme compensates by providing robust Flap 5'→3' exonuclease activity for effective TaqMan probe cleavage.
- Target Range: When using RNA as the template, the mix supports RT-PCR of fragments up to 500 bp, with optimal amplification efficiency in the range of 70–150 bp.
- Glycerol-Free Lyophilization Compatibility: The 5× CM HS-G3mTTx Mix contains no glycerol, making it compatible with our proprietary 3M Spherical Lyophilization Excipient for easy preparation of lyophilized primer-included PCR beads. This design facilitates streamlined integration of research, development, and manufacturing, while enhancing flexibility and simplifying reagent formulation and immobilization.

One unit is defined as the amount of enzyme required to incorporate 10 nmol of dNTP into acid-insoluble material in 30 minutes at 74°C.

The G3mTTx DNA/RNA qPCR Mix contains SBS Genetech’s engineered Extreme ThermoStable Reverse Transcriptase (ET RTase). This enzyme withstands 95 °C and efficiently reverse transcribes RNA up to 500 nt in length. Unlike conventional reverse transcriptases restricted to 37–50 °C, ET RTase enables cDNA synthesis at virtually any temperature, determined only by primer annealing conditions.
In addition, the system allows one-tube processing of crude samples—heat lysis, nucleic acid release, reverse transcription, and PCR amplification—streamlining workflows and redefining the format of molecular diagnostic reagents and instrumentation.
ET RTase is derived from a thermostable DNA polymerase and re-engineered using electronic scaffold reconstruction technology. Its ligand-binding core was modified to eliminate Mn²⁺ dependence, achieving strong reverse transcription activity with Mg²⁺ alone. This innovation removes the inhibitory effects of Mn²⁺ on PCR.
SBS Genetech provides two versions of the G3mTTx Mix:
CM Version – More flexible and user-friendly.
DualHS Version – Requires slightly stricter handling but delivers more rigorous amplification control.
Both formulations are glycerol-free. When combined with SBS Genetech’s proprietary 3M Spherical Lyophilization Excipient, they can be readily prepared into lyophilized beads containing amplification primers. This integrated design simplifies research, development, and production workflows while maintaining flexibility and reducing the complexity of reagent development and fixed-format manufacturing.
SBS Genetech also offers customized production of qPCR lyophilized beads, tailored to specific research or diagnostic needs.
