Description

This kit utilizes DNA binding columns and a novel unique solution system, suitable for rapidly and simply extracting genomic DNA from plant samples containing phenolics, polysaccharides, and enzyme inhibitors. Purification of DNA from one or multiple fresh samples of 100mg or 20mg of dried plant material can be completed within 30 minutes. The extraction process does not require the use of toxic organic compounds such as phenol-chloroform extraction, nor does it involve time-consuming isopropanol or ethanol precipitation. It efficiently removes impurities such as polysaccharides, phenolics, and enzyme inhibitors, allowing the purified DNA to be directly used for PCR, restriction enzyme digestion, hybridization, and other experiments.

Fresh or dried plant tissues (cells) are lysed after grinding, with proteins, polysaccharides, and cell debris precipitated and removed. Subsequently, genomic DNA selectively binds to the silica membrane within the centrifuge column under high-salt conditions, followed by a series of rapid wash and centrifugation steps to further remove polysaccharides, polyphenols, cellular metabolites, proteins, and other impurities. Finally, low-salt elution buffer is used to elute pure genomic DNA from the silica membrane.
