Acrylate‑Modified Poloxamer (F127DA)

Acrylated Poloxamer F127 (F127DA) is a photocrosslinkable and thermosensitive derivative of Pluronic F127 (Poloxamer 407), an FDA‑approved PEO–PPO–PEO triblock copolymer widely used in pharmaceutical formulations for its excellent biocompatibility, biodegradability, and self‑assembly behavior. Native F127 forms diverse nanostructures and serves as an effective solubilizer and carrier for hydrophobic drugs, transitioning from solution to physical gel at physiological temperature (≈37 °C). By introducing acrylate groups at both polymer termini, F127DA retains the thermosensitive and self‑assembling properties of Pluronic F127 while gaining rapid light‑induced crosslinking capability in the presence of photoinitiators. This enables the formation of robust, low‑swelling hydrogels with high mechanical strength, making F127DA an advanced biomaterial for drug delivery, wound healing, cell encapsulation, bio‑3D printing, microfluidic hydrogel fabrication, and composite hydrogel crosslinking.

Description

Pluronic F127, also known as Poloxamer 407, is an amphiphilic PEO–PPO–PEO triblock copolymer with excellent biocompatibility and biodegradability. It has been approved by the U.S. FDA for pharmaceutical applications. Driven by the self‑assembly behavior of its hydrophobic PPO block, F127 can form a variety of nanostructures and is widely used for solubilization, controlled release, and delivery of hydrophobic drugs. The material also exhibits pronounced thermosensitivity, forming a physical gel at physiological temperature (≈37 °C) and reverting to a solution at lower temperatures.

Our F127DA is a photosensitive biomaterial obtained by introducing acrylate groups at both ends of Pluronic F127. While retaining its inherent thermosensitive and self‑assembly characteristics, F127DA gains rapid photocrosslinking capability in the presence of photoinitiators. Upon light exposure, it forms hydrogels with excellent mechanical strength and minimal swelling at 37 °C. F127DA can also serve as a crosslinker in composite hydrogel systems. It is now widely used in advanced research fields including drug delivery, wound dressings, cell protection, bio‑3D printing, and microfluidic devices.
