Tth DNA Ligase catalyzes the formation of a phosphodiester bond between the 5’-phosphate and 3’-hydroxyl groups in double-stranded DNA, using NAD+ as a coenzyme and energy source for the reaction. Ligation occurs only when the oligonucleotide is perfectly paired with the complementary target DNA and there are no gaps between them. It is inactive on single-stranded DNA or RNA and blunt-ended DNA. Therefore, it can detect single base mutations. Its high thermal stability allows ligation under high-density hybridization conditions, enabling precise detection of SNPs with high specificity and stringency.

Description

Tth DNA Ligase catalyzes the formation of a phosphodiester bond between the 5’-phosphate and 3’-hydroxyl groups in double-stranded DNA, using NAD+ as a coenzyme and energy source for the reaction. Ligation occurs only when the oligonucleotide is perfectly paired with the complementary target DNA and there are no gaps between them. It is inactive on single-stranded DNA or RNA and blunt-ended DNA. Therefore, it can detect single base mutations. Its high thermal stability allows ligation under high-density hybridization conditions, enabling precise detection of SNPs with high specificity and stringency.

Our Tth DNA Ligase is a recombinant protein expressed and purified through multiple steps.

 

Activity Definition

One unit of activity is defined as the amount of enzyme required to ligate 50% of 1 μg of BstEII-digested λ DNA fragments (12 bp sticky ends, equivalent to 0.0338 pmole) in a 20 μl reaction system at 45°C within 15 minutes.

 

Activity Assay Conditions

  • 20 mM Tris-HCl (pH 7.6)
  • 25 mM Potassium Acetate
  • 10 mM Magnesium Acetate
  • 1 mM NAD+
  • 10 mM DTT
  • 0.1% TritonX-10
  • Reaction at 45°C
