Taq DNA Ligase catalyzes the formation of phosphodiester bonds between the 5'-phosphate end and the 3'-hydroxyl end of adjacent DNA strands. This DNA ligation reaction occurs only when two oligonucleotide chains are perfectly complementary to the target DNA and there are no gaps between the two oligonucleotide chains. It is not active on single-stranded DNA or RNA or on blunt-ended DNA. Therefore, it can be used to detect single-base mutations. Taq DNA Ligase utilizes NAD+ as a cofactor. It exhibits activity within the temperature range of 37-75°C. Its high thermal stability allows for ligation under high-stringency hybridization conditions and precise SNP detection with high specificity. Our Taq DNA Ligase is recombinantly expressed and prepared through multi-step purification of the recombinant protein.

Description

Activity Definition

One unit of activity is defined as the amount of enzyme required to ligate 50% of 1 μg of λ DNA fragments (with 12 bp sticky ends) digested with BstEII within 15 minutes under the reaction conditions of 1× Taq DNA Ligase Buffer at 45°C.

 

Activity Assay Conditions

20 mM Tris-HCl, 25 mM KAc, 10 mM Mg(Ac)2, 10 mM DTT, 1 mM NAD, and 0.1% Triton X-100 (pH 7.6 @ 25°C), incubation at 45°C.
