Description
Broadonase Salt‑tolerant Super Nuclease (≥99%), also referred to as Broadonase Salt Active Nuclease (≥99%), is a salt‑tolerant, non‑specific endonuclease engineered from Serratia marcescens and recombinantly expressed and purified from E. coli. This engineered nuclease exhibits optimal activity at high salt concentrations (500 mM) and efficiently degrades all forms of DNA and RNA—including single‑stranded, double‑stranded, linear, circular, native, or denatured nucleic acids—producing 5'-monophosphate oligonucleotides of 3–5 bases in length [1]. It is widely used for nucleic acid removal in recombinant proteins, viral vaccines, and other biological products, as well as for reducing viscosity in cell, tissue, or microbial lysates.

Features
Broadonase Salt‑tolerant Super Nuclease (≥99%) exhibits significantly enhanced salt tolerance compared with the standard Broadonase Super Nuclease. When salt concentrations exceed 100 mM, the activity of the non‑salt‑tolerant version (D7121) is markedly inhibited, often resulting in incomplete nucleic acid digestion. In contrast, the salt‑tolerant formulation achieves optimal activity at 500 mM NaCl, and even at 700 mM salt, it retains over 60% of its enzymatic activity.
This product operates on the same fundamental principles and follows usage methods comparable to NEB’s NEBExpress Salt Active Nuclease (M0764S) and Sigma’s Salt Active Nuclease (SAN) (SRE0015). In addition to the salt‑tolerant formulation, we also offer non‑salt‑tolerant Broadonase Super Nuclease (≥99%) and Broadonase Super Nuclease (≥99%, with His‑tag).
Storage
Store at −20°C. Stable for one year under recommended conditions.
Notes
- Storage at −80°C is not recommended, as repeated freeze–thaw cycles will reduce enzymatic activity.
- Broadonase Salt‑tolerant Super Nuclease (≥99%) exhibits optimal activity at a final NaCl concentration of 500 mM. The NaCl concentration in the Reaction Buffer (10×) may be adjusted according to the salt level of the sample to ensure a final reaction NaCl concentration of 500 mM.
- Magnesium ions are essential catalytic cofactors for Broadonase Salt‑tolerant Super Nuclease (≥99%). A reaction buffer containing 1–2 mM Mg²⁺ is required for enzymatic activity.
- The product is supplied at 100 U/µl. For dilution to 1 U/µl or other working concentrations, use Reaction Buffer (1×) or the enzyme storage solution.
- For research use only. Not intended for clinical diagnosis, therapeutic applications, food, or pharmaceutical use. Do not store in residential environments.
