The Tissue Single-Cell Dissociation Stop Solution is designed to terminate tissue or cell enzymatic digestion reactions, rapidly neutralizing enzyme activity and protecting cell integrity. It is widely applied in the preparation of single-cell suspensions. This solution effectively halts enzymatic reactions induced by tissue dissociation enzymes in various animal organs (such as brain, liver, spleen, kidney, lung, and tumor tissues), thereby preventing damage from over-digestion and significantly improving the success rate and data quality of downstream experiments including flow cytometry, single-cell sequencing, and cell culture.

Description

The Tissue Single-Cell Dissociation Stop Solution is designed to terminate tissue or cell enzymatic digestion reactions, rapidly neutralizing enzyme activity and protecting cell integrity. It is widely applied in the preparation of single-cell suspensions. This solution effectively halts enzymatic reactions induced by tissue dissociation enzymes in various animal organs (such as brain, liver, spleen, kidney, lung, and tumor tissues), thereby preventing damage from over-digestion and significantly improving the success rate and data quality of downstream experiments including flow cytometry, single-cell sequencing, and cell culture.

 

Features

  • Serum-free formulation: No animal serum proteins; defined composition avoids interference with downstream experiments.
  • Ready-to-use design: No preparation required; use directly after opening.
  • Broad-spectrum applicability: Contains multiple protective agents suitable for diverse tissues and organs.
  • Extended preservation: Maintains tissue integrity for up to 96 hours at 2–8 °C.

 

Protocol

  1. After enzymatic digestion is complete, immediately add an equal volume or up to twice the volume of Stop Solution to the digestion system (adjust volume according to enzyme concentration and reaction conditions).
  2. Mix gently and incubate at room temperature for 1–2 minutes to fully terminate enzyme activity.
  3. Centrifuge to collect cells at 300–400 × g for 5 minutes, discard the supernatant.
  4. Resuspend cells in pre-cooled PBS or culture medium for downstream applications (e.g., flow cytometry, single-cell sequencing, cell culture).

 

Notes

  • Mix thoroughly before use. If precipitation occurs, centrifuge and use the supernatant.
  • Digestion conditions vary by tissue type; pre-experiments are recommended to optimize Stop Solution volume and incubation time.
  • Supplied sterile; maintain aseptic conditions during use to avoid contamination.
  • For research use only.
