Description

The High-Efficiency Debris Removal Reagent is specifically designed for purifying single-cell suspensions prepared from enzymatic digestion or mechanical dissociation of various animal organs (such as brain, liver, spleen, kidney, lung, and tumor tissues). Using a unique gradient separation principle, this reagent rapidly and selectively removes cell debris, dead cells, and impurities, while retaining highly viable and pure target cells (e.g., primary cells, immune cells). It significantly enhances the success rate and data quality of downstream experiments including flow cytometry, single-cell sequencing, and cell culture.

 

Principle

This reagent is a specially formulated isotonic, sterile solution with precisely optimized density and viscosity. During centrifugation, it forms a stable density barrier:

  • High-density/large particles sediment: Cell debris, dead cells (membrane-compromised), and red blood cell aggregates quickly sediment through the barrier to form a pellet.
  • Low-density/viable cells remain suspended: Intact, viable target cells with lower density stay above the barrier in the middle-to-upper layer. By simply aspirating this layer, pure viable cells can be efficiently separated from debris.

Applications

  • Pre-treatment for flow cytometry: Removes debris, reduces non-specific background, improves sorting precision.
  • Preparation for single-cell RNA sequencing: Provides high-purity, high-viability single-cell suspensions for high-quality sequencing data.
  • Primary cell culture and functional studies: Purified cells adhere and grow more effectively, with reduced interference from impurities.
  • Functional assays (cytotoxicity, proliferation, apoptosis): Ensures accuracy and reproducibility.
  • Isolation and enrichment of rare cells (e.g., stem cells, circulating tumor cells).

 

Protocol

  1. Sample preparation: Resuspend dissociated single-cell suspension in PBS at 1–5 × 10⁶ cells/mL. Mix with reagent at a 1:1 volume ratio; increase reagent proportion if debris is abundant.
  2. Centrifugation: Spin at 400–800 × g for 10 minutes (full acceleration/brake enabled). Debris accumulates in the upper layer; intact cells sediment to the bottom.
  3. Cell collection: Carefully discard supernatant and resuspend the cell pellet in PBS.
  4. Washing/resuspension: Wash cells 1–2 times with PBS or culture medium to obtain high-purity, high-viability target cells for downstream experiments.

 

Notes

  • Debris levels vary by tissue type; optimize reagent-to-suspension ratio through preliminary experiments for best results.
  • Supplied sterile; maintain aseptic conditions during use to avoid contamination.
