Max Thermo-stable M-MLV Reverse Transcriptase (Max RTase) is derived from in vitro evolution of M-MuLV RTase. This enzyme not only possesses RNA-dependent DNA polymerase activity but also retains RNase H activity. Compared with wild-type M-MuLV RT, the engineered enzyme exhibits significantly improved thermal stability, robust amplification performance, and a high synthesis rate. Description

Max Thermo-stable M-MLV Reverse Transcriptase (Max RTase) is derived from in vitro evolution of M-MuLV RTase. This enzyme not only possesses RNA-dependent DNA polymerase activity but also retains RNase H activity. Compared with wild-type M-MuLV RT, the engineered enzyme exhibits significantly improved thermal stability, robust amplification performance, and a high synthesis rate. Max RTase enables reproducible cDNA synthesis across a broad temperature range (37–65 °C).

Thanks to its enhanced thermal stability, Max RTase maintains full activity throughout the reverse transcription reaction, yielding high amounts of cDNA and enabling synthesis of RNA transcripts longer than 6 kb. The reaction temperature can be raised to 65 °C to efficiently transcribe RNA regions with complex secondary structures. Reverse transcription can be completed within 15 to 30 minutes.

 

Unit Definition

One activity unit is defined as the amount of enzyme required to catalyze the incorporation of 1 nmol of dTTP into acid-insoluble material within 10 minutes at 37 °C, using poly(rA) as the template and oligo(dT) as the primer.

 

Storage & Transportation

  • The core enzyme product is supplied as lyophilized microbeads. When unopened, store below −15 °C (valid for 36 months).
  • After reconstitution with Storage Buffer, store below −15 °C (valid for 12 months).
  • After reconstitution with ddH₂O: store below −15 °C (valid for 2 months); store below −60 °C (valid for 12 months).
  • Shipping conditions: Ambient temperature (<37 °C) within 28 days. Upon receipt, store the product below −15 °C.
