  • What is Recombinase Polymerase Amplification (RPA)?

    A rapid, low-temperature isothermal amplification technology enabling sensitive nucleic acid detection without thermal cycling

    Recombinase Polymerase Amplification (RPA) is a rapid and highly sensitive isothermal nucleic acid amplification technology that enables DNA and RNA detection at constant temperatures, typically between 37°C and 42°C. Unlike conventional PCR, which requires repeated temperature cycling, RPA amplifies target nucleic acids under simple, low-temperature conditions without the need for complex thermal cyclers.
    By combining the activities of recombinase, single-stranded DNA binding proteins (SSB), and strand-displacing DNA polymerase, RPA enables efficient target amplification within minutes. This unique mechanism makes RPA particularly suitable for applications requiring rapid turnaround time, minimal instrumentation, and decentralized testing.
    Today, RPA has become a powerful molecular detection platform for point-of-care testing (POCT), infectious disease diagnostics, food safety testing, veterinary applications, environmental monitoring, and research workflows.

    RPA achieves rapid amplification through a unique enzyme-driven mechanism that mimics natural DNA replication processes

    Recombinase-Primer Complex Formation

    Recombinase proteins bind primers and facilitate their invasion into homologous target DNA sequences.

    Strand Separation and Target Recognition

    The recombinase-primer complex searches for complementary sequences and initiates strand exchange without high-temperature denaturation.

    DNA Extension

    A strand-displacing DNA polymerase extends the primers, generating new DNA products while continuously amplifying the target sequence.

    Signal Generation

    The amplified products can be detected through fluorescence, lateral flow assays, real-time monitoring, or integrated CRISPR-based detection systems.

    To ensure robust assay performance and flexibility, our RPA system is supported by a comprehensive enzyme platform, including key functional proteins such as T4 UvsX Recombinase, T4 UvsY Protein, T4 Gene 32 Protein, Bsu DNA Polymerase, Sau DNA Polymerase, Exonuclease III, Bst DNA/RNA Polymerase, and Endonuclease IV, which collectively enable efficient strand invasion, DNA synthesis, and reaction control in isothermal amplification systems.
