A strand of DNA is used as a template for synthesizing a strand of RNA by RNA polymerase (e.g. T7 Thermostable RNA Polymerase). During this process, RNA polymerase binds to a site called the promoter and starts copying the nucleotides, where deoxyribonucleotides are replaced by ribonucleotides.
Complementary DNA (cDNA) is generated from an RNA template by reverse transcriptase (e.g. Ultra Thermo-stable M-MLV Reverse Transcriptase). During this process, RNasin is usually used to prevent the degradation of RNA template.
