Description
Faustovirus Capping Enzyme (FCE) is a single-subunit mRNA capping enzyme derived from the Faustovirus S17 strain. FCE possesses the three enzymatic activities required for adding a Cap-0 structure to mRNA: Triphosphatase (TPase), Guanylyltransferase (GTase), and (Guanine-N7)-Methyltransferase (N7 MTase). It catalyzes the addition of an N7-methylguanosine cap structure (m7GpppNp) to the 5' end of eukaryotic mRNA at the triphosphate and diphosphate positions, generating mRNA with a Cap-0 structure.
Compared to Vaccinia Capping Enzyme (VCE), FCE exhibits a broader reaction temperature range and higher enzymatic activity. It retains capping activity at 37°C and up to 55°C, making it highly adaptable. Additionally, FCE can work synergistically with mRNA Cap 2'-O-Methyltransferase to directly add a Cap-1 structure to mRNA in a single step.
The addition of Cap-0 and Cap-1 structures, along with poly(A) tailing, is crucial for mRNA maturation. The Cap-0 structure protects mRNA from 5'→3' exonuclease degradation, enhancing its stability. Furthermore, it prevents triphosphorylated mRNA from triggering the body's innate immune response.

One unit of Faustovirus Capping Enzyme is defined as the amount of enzyme required to convert 75 pmol of a 20-mer transcript to Cap-0 RNA within 30 minutes at 37°C.

Inactivation or Inhibition
Faustovirus Capping Enzyme is inactivated by heating at 70°C for 10 minutes.
Storage
The minimum shelf life is 2 years at -20°C.
Precautions
- Keep the product on an ice box or in an ice bath during use. Immediately store at -20°C after use.
- Since RNA handling is involved, strictly follow RNA operation protocols to avoid RNase contamination.
- Use RNase-free consumables such as pipette tips and centrifuge tubes, or treat them with DEPC to ensure RNA integrity.
- Select appropriate handling methods based on the specific application. Additional reagents such as RNase Inhibitor and ultrapure water may be required.
- SAM is unstable at pH 7–8 and 37°C. Prepare a fresh working solution based on the actual reaction setup before starting and keep it on ice to prevent degradation.
