Description

Tris-Glycine (referred to as TG below) is a white to pale yellow granule. Each pouch of TG instant granules can be used to prepare 1 L of 1× TG buffer solution, making it easy and convenient to use. The main components of TG are Tris and glycine, with a concentration of 25 mM Tris and 192 mM glycine in 1× TG buffer solution.

1. TG buffer solution, when prepared as 1× in 20% ethanol, is commonly used for rapid Western Blotting and is primarily used in wet transfers.
2. It is also used for preparing non-denaturing protein polyacrylamide gel electrophoresis (Native-PAGE) running buffer.

Components

• Tris-Glycine: 50 Pouches (50×1 L)

Protocol

1. Measure approximately 600 ml of distilled water and add it to a beaker (if it's intended for membrane transfer in immunoblotting, add 200 ml of ethanol and approximately 400 ml of distilled water). Place a magnetic stir bar into the beaker.

2. Position the beaker on a magnetic stirrer and slowly add the entire contents of 1 pouch of TG instant granules. Stir the solution until it is completely dissolved.

3. Add distilled water to the TG solution from step 2, filling it up to 1 L, thus obtaining a 1× solution.
