Description
Thermostable dsAP Nuclease is a heat-stable endonuclease derived from thermophilic bacteria. It specifically recognizes abasic (AP) sites in double-stranded DNA and cleaves the first phosphodiester bond at the 5' end of the AP site, generating a 3' hydroxyl and a 5' deoxyribose phosphate (dRP) terminus.
This enzyme is active exclusively on AP-containing double-stranded DNA (dsDNA) and exhibits no activity on single-stranded DNA (ssDNA), dsDNA without AP sites, or AP-containing ssDNA.
Its optimal activity temperature is 60–70°C, with progressive inactivation occurring above 85°C.
Unit Definition
One unit is defined as the amount of enzyme required to cleave 1 pmol of AP-containing double-stranded oligonucleotide in a 10 µl reaction system at 60°C within 15 minutes.
