Description
Efficient translation of most eukaryotic mRNAs requires a 7-methylguanosine cap at the 5’ end and a Poly(A) tail at the 3’ end. The T7 ARCA in vitro mRNA Synthesis Kit enables the rapid synthesis of fully capped and tailed mRNA in vitro.
During transcription, T7 RNA polymerase incorporates an anti-reverse cap analog (ARCA) at the mRNA’s 5’ end. After a brief incubation with DNase I, the template DNA is removed, followed by Poly(A) polymerase extension to add a Poly(A) tail to the capped mRNA.
The resulting RNA is suitable for a wide range of downstream applications, including RNA structure and function studies, nucleic acid enzyme biochemistry research, RNase protection assays, blot hybridization, antisense RNA and RNA interference (RNAi) experiments, microarray analysis, microinjection, in vitro translation, and RNA vaccine development.
T7 RNA polymerase recognizes the T7 promoter sequence (TTCTAATACGA CTCACTATAGG) and initiates transcription from the -2 position G base. This kit enables high-efficiency transcription from minimal sample input, yielding over 30 μg of mRNA per reaction, with transcription lengths exceeding 4000 nucleotides. Over 80% of the transcription products contain a 7-methylguanosine cap, and the Poly(A) tail length exceeds 80 nucleotides.
