The basic principle of Multiple Displacement Amplification (MDA) relies on random hexamer primers and phi29 DNA Polymerase to carry out amplification under isothermal conditions. During the reaction, the random hexamers anneal arbitrarily to the DNA template, after which phi29 DNA Polymerase initiates synthesis simultaneously at multiple loci across the genome. As replication progresses, the newly synthesized DNA strands displace the complementary strands of the template, which in turn serve as fresh templates for further rounds of amplification. Capable of yielding high-mass, high-quality genomic DNA from trace biological samples, MDA has emerged as the most widely adopted method for single-cell whole genome amplification (WGA).

Storage
