Utilizing S-adenosylmethionine (SAM) as a methyl donor, it transfers a methyl group to the 2'-O position of the first nucleotide adjacent to the Cap-0 structure (m7GpppNp) at the 5' end of the mRNA, resulting in the formation of Cap-1-modified mRNA (m7GpppNmp). This modification enhances mRNA stability and translation efficiency, improving expression levels after microinjection and transfection. Additionally, Cap-1 helps mRNA evade certain innate immune responses in cells.

Description

mRNA Cap 2'-O-Methyltransferase (mRNA 2'-O-MTase) is an mRNA capping enzyme that converts the Cap-0 structure into a Cap-1 structure. Utilizing S-adenosylmethionine (SAM) as a methyl donor, it transfers a methyl group to the 2'-O position of the first nucleotide adjacent to the Cap-0 structure (m7GpppNp) at the 5' end of the mRNA, resulting in the formation of Cap-1-modified mRNA (m7GpppNmp). This modification enhances mRNA stability and translation efficiency, improving expression levels after microinjection and transfection. Additionally, Cap-1 helps mRNA evade certain innate immune responses in cells.

mRNA Cap 2'-O-Methyltransferase specifically targets N7-methylguanosine-capped mRNA (m7GpppNp), meaning it requires an existing Cap-0 structure for activity. It does not modify mRNA with 5' terminal structures such as pN, ppN, pppN, or GpppN. Typically, Faustovirus Capping Enzyme is used to generate Cap-0 mRNA, followed by mRNA Cap 2'-O-Methyltransferase to introduce the Cap-1 modification. Alternatively, both enzymes can act synergistically to generate Cap-1 mRNA in a single step.

Produced by Escherichia coli (E. coli) expression of the vaccinia virus mRNA Cap 2'-O-Methyltransferase gene, followed by purification.

 

Activity Unit Definition

One unit of mRNA Cap 2'-O-Methyltransferase is defined as the amount of enzyme required to methylate 10 pmoles of an 80-nt-long capped RNA transcript in 1 hour at 37ºC.

 

Purity

  • Free from DNA endonucleases and exonucleases.
  • Free from RNases.
  • Free from phosphatases.

 

Inactivation/Inhibition

Heating at 70ºC for 10 minutes will inactivate mRNA Cap 2'-O-Methyltransferase.

 

Storage

The minimum shelf life is 2 years at -20°C.

 

Precautions

  • During use, keep the product on an icebox or ice bath, and return it to -20ºC storage immediately after use.
  • Since RNA manipulation is involved, strict RNA-handling protocols must be followed to prevent RNase contamination.
  • Laboratory consumables such as pipette tips and centrifuge tubes must be RNase-free or treated with DEPC.
  • Depending on the specific application, additional reagents such as RNase Inhibitor and ultrapure water may be required.
  • S-adenosylmethionine (SAM) is unstable at pH 7-8 and 37ºC. Prepare a fresh working solution immediately before use and keep it on ice to prevent degradation.
  • For research use only: This product must not be used for clinical diagnosis, treatment, food, or pharmaceuticals, and must not be stored in residential settings.
  • Safety precautions: Wear a lab coat and disposable gloves while handling the product.
