Description
Enhanced Adeno-associated Virus Cell Lysis Buffer Plus (10×) is a chemically defined (CD), animal-origin-free (AOF) lysis buffer specially formulated to thoroughly lyse AAV-packaging cells such as HEK293 and maximize adeno-associated virus (AAV) recovery. Formulated with mild detergents, this reagent enables efficient cell lysis and complete AAV release without supplemental mechanical homogenization, delivering a gentle, user-friendly and high-efficiency workflow for intracellular AAV purification.
Adeno-associated Virus (AAV), a small single-stranded DNA virus belonging to the genus Dependovirus in the family Parvoviridae, encompasses over 100 serotypes. Upon cellular transduction, AAV rarely integrates into the host genomic DNA yet supports long-term stable transgene expression, notably in a broad range of non-dividing cell types. Featuring high viral titers, minimal in vivo immunogenicity following parenteral administration, and robust infectivity toward both dividing and quiescent cells, AAV has gained extensive applications across biomedical research.
Composed of a single formulation component, this buffer requires only one dilution step prior to use, streamlining cell lysis and subsequent AAV extraction.
The product ensures straightforward handling and excellent experimental reproducibility. Simply mix the stock buffer with cell suspension at a volumetric ratio of 1:9, followed by 2-hour incubation with shaking at 37°C to achieve effective cell lysis and AAV liberation. It outperforms the labor-intensive freeze-thaw cycling method with superior ease of operation and consistent assay performance.
This buffer delivers markedly improved AAV harvesting efficiency. Under identical experimental conditions, its AAV yield is approximately 114% higher than that obtained via repeated freeze-thaw treatment, enabling substantial enhancement of overall AAV production output.
This product is free of Triton X-100, which better preserves the structural integrity and infectivity of AAV particles.
For AAV samples harvested with this lysis buffer, viral titer quantification is recommended using the Universal AAV Titration Kit by qPCR.
Storage
Store at −20°C for up to 12 months; stored at 4°C, the product remains stable for a minimum of one week.

- To guarantee optimal performance, avoid repeated freeze-thaw cycles of Enhanced Adeno-associated Virus Cell Lysis Buffer Plus (10×). Aliquot the buffer appropriately prior to use as needed.
