DNA-Free HS Pfu DNA Polymerase

DNA-Free HS Pfu DNA Polymerase is a hot-start Pfu DNA polymerase derived from recombinant expression in Escherichia coli of Pyrococcus furiosus DNA polymerase, with a molecular weight of 90 kDa. Its fidelity is approximately 18 times higher than that of ordinary Taq DNA polymerase. This enzyme has been specially modified to possess strong amplification ability and extension speed, with an amplification fragment length of up to 5-6 kb and an extension speed of 1 kb/min (at 72℃). It exhibits 5’→3’ polymerase activity and 3’→5’ exonuclease activity. The amplification products have blunt ends and are not suitable for TA cloning.

Description

DNA-Free HS Pfu DNA Polymerase is a hot-start Pfu DNA polymerase derived from recombinant expression in Escherichia coli of Pyrococcus furiosus DNA polymerase, with a molecular weight of 90 kDa. Its fidelity is approximately 18 times higher than that of ordinary Taq DNA polymerase. This enzyme has been specially modified to possess strong amplification ability and extension speed, with an amplification fragment length of up to 5-6 kb and an extension speed of 1 kb/min (at 72℃). It exhibits 5’→3’ polymerase activity and 3’→5’ exonuclease activity. The amplification products have blunt ends and are not suitable for TA cloning.

Our DNA-Free HS Pfu DNA Polymerase, on the other hand, is devoid of genomic and plasmid DNA from the production strain. This makes DNA-free Pfu particularly suitable for amplifying, sequencing, and cloning 16S and 23S rDNA genes, as well as for standard applications like PCR and primer extension.

 

Unit Definition

The amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide into acid-insoluble material within 30 minutes at 72℃ using salmon sperm DNA as the template/primer is defined as one unit of activity (U).

 

Features

  • Completely free from bacterial DNA.
  • This enzyme is a hot-start polymerase, which improves amplification specificity and reduces non-specific amplification and primer dimer formation.
  • Specially modified compared to traditional Pfu DNA polymerase, it has enhanced amplification ability and extension speed, capable of effectively amplifying 5-6 kb DNA fragments under 1 kb/min extension conditions.
  • Good thermal stability: Half-life exceeds 60 minutes at 98℃.
  • Highest fidelity: 10 times the fidelity of Taq DNA polymerase.
  • Not tolerant to dUTP and dITP.
  • PCR products have blunt ends and can be directly used for blunt-end cloning.

  • Preparation of PCR products for gene cloning or protein expression.
  • Blunt-end PCR cloning (removing 3’ overhangs).
  • Site-specific mutagenesis.
