Description
BCA Peptide Quantitation Assay Kit (Colorimetric), also known as the BCA Peptide Quantitation Assay Kit (Colorimetric) or the BCA Quantitative Colorimetric Peptide Assay Kit, is developed based on the BCA method—one of the most commonly used methods worldwide for protein concentration detection. This kit provides improved BeyoBCA reagents and special chelating agents, along with high-quality peptide standards. It offers simple, stable, highly sensitive, and compatible peptide concentration measurement, making it suitable for subsequent applications such as quantification, LC, MS, and LC/MS, as well as detecting recovery rates after peptide purification.

Features
- This kit is suitable for detecting peptide concentrations with high sensitivity. BCA Protein Quantification Assay Kit and Enhanced BCA Protein Assay Kit are used for detecting undigested proteins. In contrast, this kit is designed for fully digested peptides. Compared to using the BCA Protein Quantitation Assay Kit for peptide detection, this kit offers more than double the sensitivity, with a detection limit as low as 5µg/ml and a minimum detectable peptide amount of 0.1µg. The sample volume required is 1-20µl.
- Fast Detection: The detection speed is fast, typically requiring only 15 minutes of incubation before absorbance measurement.
- The kit provides a good linear relationship within the concentration range of 0-1000µg/ml.
This kit demonstrates excellent chemical compatibility. It can be used to measure peptide concentrations without being affected by most chemicals present in samples. However, it is influenced by chelating agents and slightly higher concentrations of reducing agents. Ensure EDTA is below 5mM, and avoid DTT (Dithiothreitol) and TCEP (Tris(2-carboxyethyl)phosphine). This kit is compatible with amine-reactive labeling reagents (e.g., biotinylation reagents). The detailed compatibility of various substances in samples with this kit is as follows:

This kit is not recommended for quantifying peptides containing only a single type of amino acid. Factors such as peptide hydrophobicity, length, structure, and specific amino acid content may influence the final results. If the kit is unsuitable, or if the sample quantity is limited or at low concentration, the fluorometric Peptide Quantitation Assay Kit (P0396) is recommended due to its higher sensitivity compared to the colorimetric method. The fluorometric method has a detection limit of 2µg/ml, with a minimum detectable peptide amount of 0.02µg and a required sample volume of only 1-10µl. The small package of this kit can test 100 samples, and the medium package can test 500 samples.
Storage
- Store at 4ºC for up to one year.
- BCA Reagent B should be stored protected from light.
- After opening, store Peptide Standard (1mg/ml) at -20ºC.
Precautions
- If Reagents A, B, or C precipitate during long-term storage, dissolve the precipitate by heating or stirring in a 37ºC water bath. If turbidity persists, mix thoroughly until a clear solution is obtained.
- A microplate reader is required to measure absorbance at 480nm, using a 96-well plate. If a microplate reader is not available, a standard spectrophotometer can be used. Ensure the working solution volume is sufficient to meet the minimum detection volume of the cuvette. Adjust the sample and standard volumes accordingly. Note that using a spectrophotometer may significantly reduce the number of samples that can be tested per kit.
- If samples contain a high amount of undigested proteins, have high background due to the sample diluent or lysis buffer, or contain EDTA concentrations above 5mM or reducing agents such as DTT or TCEP, it is recommended to use Peptide Quantitation Assay Kit (Fluorometric).
- Allow all reagents to equilibrate to room temperature before use. Immediately store reagents under the conditions specified in the kit after use.
- This product is intended for scientific research use by professionals only. It is not for clinical diagnosis or treatment, nor for use in food or medicine. Do not store it in common household environments.
