Description
Peptide Quantitation Assay Kit (Fluorometric), also known as the Quantitative Fluorometric Peptide Assay Kit, is designed to quantify peptide concentration using an amine-reactive fluorescent reagent that specifically labels the amino terminus of peptides. Fluorescence is detected at an excitation wavelength of 390nm and an emission wavelength of 475nm. This kit provides high-quality peptide standards, achieving simple, stable, highly sensitive, and compatible peptide concentration measurement. It is suitable for subsequent applications such as quantification or LC, MS, and LC/MS, as well as detecting recovery rates after peptide purification.
Components
- Reagent A
- Reagent B
- Peptide Standard (1mg/ml)
Features
- This kit is suitable for detecting peptide concentrations with high sensitivity. Compared to the BCA Peptide Quantitation Assay Kit (Colorimetric), it offers higher sensitivity, with a detection limit as low as 2µg/ml and a minimum detectable peptide amount of 0.02µg. The sample volume required is 1-10µl.
- The detection speed is faster compared to BCA Peptide Quantitation Assay Kit (Colorimetric), requiring only about 5 minutes for fluorescence measurement.
- The kit provides a good linear relationship within the concentration range of 0-1000µg/ml.
This kit demonstrates good chemical compatibility. It can be used to measure peptide concentrations without being affected by most chemicals present in samples. However, it is influenced by amine-containing buffers, so ensure ammonium bicarbonate (NH4HCO3) is below 50mM and no other free amines or ammonium salts are present. The detailed compatibility of various substances in samples with this kit is as follows:

This kit can be used for quantifying peptides composed of different amino acids as well as peptides containing only a single type of amino acid. However, factors such as peptide hydrophobicity, length, structure, specific amino acid content, or sequence may influence the final detection results. This kit is not suitable for measuring the concentration of peptides labeled with amine-reactive reagents (e.g., biotinylation reagents). If unsuitable, it is recommended to use the BeyoBCA Peptide Quantitation Assay Kit (colorimetric method) (P0397). The main features and differences between the colorimetric and fluorometric peptide quantitation assay kits are summarized below:

The small package of this kit can test 100 samples, and the medium package can test 500 samples.
Storage
Store at 4ºC for up to one year. Reagent B should be stored protected from light. Storing at -20ºC extends the shelf life.
Precautions
- If Reagents A and B precipitate during long-term storage, dissolve the precipitate by heating or stirring in a 37ºC water bath before use.
- A fluorescence microplate reader capable of detecting excitation at 390nm and emission at 475nm is required.
- If a larger assay volume is needed, maintain the volume ratio of Sample : Reagent A : Reagent B at 1:7:2 to avoid changes in pH or dilution of the assay reagents. Note that using larger assay volumes will significantly reduce the number of samples that can be tested per kit.
- If the sample contains more than 50mM ammonium bicarbonate (NH4HCO3) or other ammonium salts, free amines, or is labeled with amine-reactive reagents, it is recommended to use BCA Peptide Quantitation Assay Kit (Colorimetric).
- Allow all reagents to equilibrate to room temperature before use, and store them under the conditions specified in the kit immediately after use.
