dCas9 NLS (Lyophilized)

dCas9 NLS is a nuclease with a Nuclear Localization Signal (NLS) that specifically binds double-stranded DNA under the guidance of gRNA without cutting it. dCas9 NLS is a D10A/H840A double mutant created by amino acid mutations in the RuvC and HNH domains of the Cas9 nuclease. Like the wild-type Cas9 nuclease, dCas9 NLS targets the DNA sequence with the help of gRNA. However, due to mutations in the RuvC and HNH domains, dCas9 NLS has gRNA-guided DNA binding activity without endonuclease activity. Studies have shown that when dCas9 NLS is combined with proteins of different functions, it can effectively inhibit or promote gene expression. LwaCas13a Nuclease (Lyophilized)

LwaCas13a (also known as C2c2) is a crRNA-guided RNA endonuclease dependent on crRNA alone, originating from the Leptotrichia wadei strain. It can specifically recognize and cleave target single-stranded RNA, with relatively lenient requirements for the PFS sequence. Additionally, Cas13a exhibits trans-cleavage activity (i.e., collateral cleavage activity). Upon formation of a ternary complex comprising LwaCas13a protein, crRNA, and target RNA, the enzyme is activated to cleave non-specific sequences of single-stranded RNA in the system, breaking down any sequence of single-stranded RNA. BrCas12a Nuclease (Lyophilized)

When guided by RNA, Cas12b recognizes and cleaves the target DNA, activating its "collateral cleavage" activity, efficiently cleaving nonspecific DNA in the system. By designing Reporter DNA labeled with fluorescent groups or other markers at both ends, CRISPR/Cas12b can detect and amplify signals of DNA templates. The results can then be observed using a fluorometer or test paper. It is characterized by its tolerance to high temperatures of 65°C and can be used in conjunction with LAMP isothermal amplification.
