Yeast tRNA (Carrier RNA)

Yeast tRNA (Carrier RNA) is derived from brewer's yeast. It can serve as a co-precipitant for nucleic acid precipitation and as a blocking agent for RNA probe hybridization reactions (such as in situ hybridization, Northern blotting, and dot blotting). However, it is not recommended to use yeast tRNA for subsequent polynucleotide kinase or terminal transferase reactions, as yeast tRNA may compete with the substrate for enzyme activity. Although it cannot be used in reactions inhibited by exogenous RNA, as a co-precipitant, it is of high quality and low cost.

Description

Yeast tRNA (Carrier RNA) is derived from brewer's yeast. It can serve as a co-precipitant for nucleic acid precipitation and as a blocking agent for RNA probe hybridization reactions (such as in situ hybridization, Northern blotting, and dot blotting). However, it is not recommended to use yeast tRNA for subsequent polynucleotide kinase or terminal transferase reactions, as yeast tRNA may compete with the substrate for enzyme activity. Although it cannot be used in reactions inhibited by exogenous RNA, as a co-precipitant, it is of high quality and low cost. tRNA is a specific transport RNA involved in protein biosynthesis, ensuring accurate transfer of genetic information from nucleic acids to proteins. Each amino acid has more than one corresponding tRNA, with approximately 40-70 different tRNAs in each cell, consisting of 72-93 nucleotides, 5%-20% of which are rare bases, with an average molecular weight of 25,000 and a sedimentation coefficient of about 4S. While the primary structures of different tRNAs vary, they share a common cloverleaf secondary structure. The amino acid arm 3’-C-C-A-OH is the site for amino acid binding, where the amino acid's acyl group attaches to the 3’-OH of the corresponding tRNA’s adenine under the action of specific aminoacyl-tRNA synthetases. The anticodon loop, consisting of three consecutive nucleotides, forms an anticodon that complements the codon on the mRNA in the ribosome, ensuring accurate amino acid incorporation into the polypeptide chain. This codon-anticodon-amino acid correspondence allows tRNA to play a crucial role in accurately transferring information from nucleic acids to proteins.

 

Quality Control

  • Appearance: White to gray-brown powder.
  • Electrophoresis Test: Product is qualified.
  • Content: 15-20 A260 units/mg
  • Non-Specific Nuclease Endonuclease Activity Residual Detection: Product is qualified.
  • Nucleic Acid Exonuclease Activity Residual Detection: Product is qualified.
  • RNase Activity Residual Detection: Product is qualified.

 

Application

Nucleic Acid Precipitation: Adjust the solution’s monovalent cation concentration (e.g., 0.5 M NH4OAc, 0.25 M NaCl, or 0.3 M NaOAc). Add yeast tRNA to a final concentration of 10-20 μg/mL. Mix thoroughly, then add 2 volumes of ethanol. Freeze at -20°C or lower for at least 15 minutes. Centrifuge at ≧10,000×g for at least 15 minutes. Carefully remove the supernatant and resuspend the pellet in an appropriate buffer.

Note: When using yeast tRNA as a carrier and isopropanol instead of ethanol, a small amount of nucleic acid may not quantitatively precipitate. For ethanol precipitation of end-labeled oligonucleotides (e.g., 35-mers), linear polyacrylamide or glycogen are more effective co-precipitants than yeast tRNA.

 

Handling Instructions

RNA is highly sensitive to degradation by exogenous ribonucleases introduced during handling. When using this product, please wear gloves. Use RNase-free reagents, tubes, and pipette tips.

 

Thawing Instructions

Thaw completely at temperatures below 37°C. After melting, vortex for a few seconds and then place on ice. If necessary, aliquot RNA to minimize freeze-thaw cycles (≦5).

 

Storage

Store at 2-8°C.

 

Note

  • This product should be avoided from repeated freeze-thaw cycles. It is recommended to aliquot appropriately upon initial use and store in a sealed manner at -20°C or lower temperatures.
  • Handle with care and avoid contamination. Please use RNase-free tips and EP tubes. It is recommended to wear a disposable mask while performing the procedure.
  • For samples with a low quantity or when extracting viral RNA, it is advisable to use Carrier RNA. If a larger amount of RNA is expected, the addition of Carrier RNA can be chosen according to the needs.
  • Due to the presence of Carrier RNA, the concentration of viral RNA may be inaccurate. It is recommended to use qRT-PCR for quantification.
  • This product is intended for scientific research purposes by professionals only and should not be used for clinical diagnosis or treatment. It should not be used for food or drugs, and should not be stored in residential areas.
  • For your safety and health, please wear a lab coat and disposable gloves while handling.
