Description

T4 RNA Ligase is an ATP-dependent enzyme that catalyzes the formation of phosphodiester bonds between the 5’-P end and the 3’-OH end of single-stranded RNA, single-stranded DNA, or single nucleotides, either intermolecularly or intramolecularly.

T4 RNA Ligase is primarily used for ligating RNA to RNA, requiring the presence of a 5’ phosphate group and a 3’ hydroxyl group. It can ligate RNA molecules to each other and also circularize RNA molecules (as short as 8 bases).

T4 RNA Ligase can be used for ligating RNA to single nucleotides, which must be phosphorylated at both the 5’ and 3’ ends, commonly used for 3’ end labeling of RNA.

T4 RNA Ligase can also ligate DNA to RNA. When DNA provides the 5’ phosphate group and RNA provides the 3’ hydroxyl group, the ligation efficiency is high; however, when DNA provides the 3’ hydroxyl group and RNA provides the 5’ phosphate group, the ligation efficiency is very low.

T4 RNA Ligase can ligate DNA to DNA, but the efficiency is very low. It is mainly used for circularizing DNA, such as in 5’ RACE for cDNA circularization. Although DNA-to-DNA ligation is possible, it is quite challenging.

 

Application

3’ end labeling of RNA; RNA-to-RNA ligation; circularization of oligonucleotides; tRNA modification; ligation of oligonucleotides to single-stranded cDNA in 5’ RACE; introduction of non-natural amino acids at specific protein sites.

 

Source

Expressed in Escherichia coli, with the gene sourced from T4 bacteriophage.

 

Activity Definition

One unit is defined as the amount of enzyme required to catalyze the formation of 1 nmol of phosphatase-resistant circular product from 5’-[32P]-(A)12-18 in 30 minutes at 37ºC.

 

Purity

Free from endonucleases, exonucleases, RNases, and phosphatases.

 

Inactivation or Inhibition

Heating at 70ºC for 10 minutes will inactivate T4 RNA Ligase.
