Recombinant Kex2 Protease

Kex2 protease is a calcium-dependent serine protease derived from yeast. It specifically recognizes and cleaves peptide bonds at the carboxyl terminus of double basic amino acids such as Arg-Arg, Lys-Arg, and Pro-Arg. Unlike pancreatic proteases, Kex2 does not recognize and cleave peptide bonds at the carboxyl terminus of single basic amino acids such as arginine or lysine. In yeast, Kex2 protease is responsible for processing the precursors of killer toxin and α-factor. The activity of Kex2 protease is not inhibited by conventional serine protease inhibitors such as antipain, PMSF, or TPCK. The recombinant Kex2 protease is expressed and produced by Pichia pastoris and exhibits the same enzyme specificity as native Saccharomyces cerevisiae Kex2 enzyme. It exhibits optimal activity at pH 9.0 and is stable at pH 5.0-6.0.

Description

Cleavage of secreted peptides in yeast heterologous protein expression.

 

Protein Information

  • Unit definition: One unit of enzyme activity is defined as the amount of enzyme that catalyzes the release of 1 μmol of p-nitroaniline from the substrate Boc-QRR-pNA per minute at 25°C in a reaction system of 3 mL containing 50 mM Tris-HCl, 2 mM CaCl2, pH 8.0.
