Description
Mycoplasma Prophylactic and Elimination Reagent I is a reagent used to prevent and remove mycoplasma contamination, which is widespread in cultured cells. This reagent can prevent mycoplasma contamination and also remove existing mycoplasma contamination in cells. It mainly contains fluoroquinolone antibiotics and macrolide antibiotics.
Mycoplasma is the smallest and simplest prokaryote. Mycoplasmas have the following characteristics: they lack a cell wall structure, so many common antibiotics targeting the cell wall, such as penicillin or β-lactam antibiotics, are ineffective against them; their size is between bacteria and viruses, approximately 0.2-0.8 μm, so some mycoplasmas can pass through a 0.22 μm filter, making conventional filtration ineffective; many mycoplasmas rely on the host for nutrients due to their limited biosynthetic capabilities, often attaching to or dispersing among cell surfaces and between cells. These characteristics pose a risk of mycoplasma contamination during cell culture, and mycoplasma contamination has become a global issue.
Mycoplasma contamination can severely affect cell status, altering gene expression and metabolic characteristics, leading to slowed cell growth, abnormal differentiation, and cell death, which seriously impacts cell function. These factors can significantly affect the reliability and reproducibility of experimental results, making the prevention and removal of mycoplasma contamination crucial.
This mycoplasma prophylactic and elimination reagent contains two effective antibiotics against mycoplasmas: fluoroquinolone antibiotics and macrolide antibiotics. Fluoroquinolone antibiotics inhibit DNA replication by affecting the replication fork, while macrolide antibiotics inhibit protein synthesis by interfering with ribosomal translocation. This reagent is effective only against mycoplasmas and bacteria, with no impact on eukaryotic cells, thus not affecting the normal growth and function of eukaryotic cells.

Mycoplasma Removal Reagent and Mycoplasma Removal Reagent Plus are both single antibiotics in powder form. They are slightly inconvenient to use, but you can measure out small amounts according to your needs. If stored at -20°C, the powder products can last longer. Additionally, if resistance develops to a single antibiotic, it is relatively simple to switch to another antibiotic, which usually yields better results.
Mycoplasma Prophylactic and Elimination Reagent I and Mycoplasma Prophylactic and Elimination Reagent II are pre-prepared solutions, making them more convenient to use, but they must be stored at -20°C. Since these two reagents are combination antibiotics, they generally have a stronger effect in inhibiting or removing mycoplasma. However, if resistance develops, subsequent handling can be more troublesome, and it is more likely that mycoplasma resistance will persist even after switching to another prevention and removal reagent.
This product is easy to use. Simply add it directly to the culture medium at a dilution ratio of 1:500 (for prevention) or 1:100 (for removal). Each milliliter of this product can prepare 500ml of prevention culture medium or 100ml of removal culture medium.
Storage
Store at -20°C, valid for two years.
Precautions
- Avoid repeated freezing and thawing of this reagent. If used multiple times, please consider appropriate aliquoting.
- If crystals or precipitates are observed after thawing, it does not affect the usage. Shaking can help dissolve the crystals or precipitates.
- It is recommended to regularly test for mycoplasma using mycoplasma detection kits, such as our Luminescence Mycoplasma Detection Kit, Mycoplasma PCR Detection Kit, or Mycoplasma Staining Detection Kit.
- This reagent can effectively inhibit or remove various mycoplasmas, but it cannot guarantee the inhibition or removal of all types of mycoplasmas encountered in actual experimental operations. Although the likelihood of mycoplasma developing resistance to this reagent is low, long-term use may still result in resistance. If the reagent is not effective, consider using our other mycoplasma removal reagents.
